Bread wheat Paragon was crossed to durum wheat Kronos mutant line T4-2235 which carries a splice acceptor site mutation in the A genome homoeologue of the TaGW2 gene (mutation G2373A). After the initial cross, four backcrosses were performed selecting progenies carrying the mutant gw2-a1 allele. The lines were then self-pollinated and homozygous mutant and homozygous wild type sibling lines were selected.
The BC4F3 sibling lines were grown in replicated glasshouse and field trials in the UK (Norwich) and US (Davis) and evaluated for grain weight and size. Lines carrying the gw2-a1 mutant allele had significantly increased thousand grain weight due to both wider and longer grains when compared to the isogenic sibling lines carrying a wild type allele at GW2-A1.
